چکیده

Leonard, C. Gomez (U.S. Army Biological Laboratories, Fort Detrick, Frederick, Md.), D. K. Mattheis, M. J. Mattheis, and R. D. Housewright. Transformation to prototrophy and polyglutamic acid synthesis in Bacillus licheniformis. J. Bacteriol. 88:220-225. 1964.-Growth conditions necessary for transformation to prototrophy of 14 auxotrophs of Bacillus licheniformis were studied. The unexpected finding of different growth requirements by each auxotroph for the development of transformable cells is discussed. Under optimal growth conditions for a serine-deficient mutant transformation frequencies of 0.1% were obtained. A medium for transformation of competent cells in liquid suspensions is described. In addition, this report presents evidence for the transformation of three nonencapsulated mutants of B. licheniformis for the ability to synthesize polyglutamic acid (capsular material).

Conversion of the nitrogen content in liquid manure into biomass and polyglutamic acid by a newly isolated strain of Bacillus licheniformis.

Extensive spreading of liquid manure onto agricultural fields causes eutrophication of ground and surface water and also pollution of the atmosphere due to the high ammonium nitrogen content. A poly(gamma-glutamic acid) (PGA)-producing strain of Bacillus licheniformis was isolated in this study and investigated for its ability to reduce the ammonium nitrogen by converting ammonium into biomass ...

Use of Response Surface Methodology to Optimize Culture Medium for Production of Poly-γ-glutamic Acid by Bacillus licheniformis

Response surface methodology (RSM) was employed to study the effects of four medium components (L-glutamic acid, citric acid, glycerol and NH4Cl) on the production of poly-γ-glutamic acid (PGA) in flask cultures by Bacillus licheniformis CCRC 12826 (ATCC 9945) with initial medium pH 6.5. The optimal PGA yield (35.34 g/L, average of four repeats) was appeared at the region where the respective c...

Isolation and molecular characterization of chitinase-deficient Bacillus licheniformis strains capable of deproteinization of shrimp shell waste to obtain highly viscous chitin.

Proteolytic but chitinase-deficient microbial cultures were isolated from shrimp shell waste and characterized. The most efficient isolate was found to be a mixed culture consisting of two Bacillus licheniformis strains, which were first determined microscopically and physiologically. Molecular characterization was carried out by sequencing the 16S rRNA gene of both strains. According to the re...

Effects of pH and aeration on gamma-poly(glutamic acid) formation by Bacillus licheniformis in controlled batch fermentor cultures.

Bacillus licheniformis ATCC 9945A was grown on Medium E in batch fermentations in which the pH was maintained at 5.5., 6.5, 7.4, and 8.25. The effects of pH on cell growth, carbon source utilization, and gamma-polyglutamic acid (gamma-PGA) production, molecular weight, and polymer stereochemistry were determined. The gamma-PGA yield was highest (15 g/L, 96 h growth time) at pH 6.5. The increase...

Conditions for competence in the Bacillus licheniformis transformation system.

Some variables in the Bacillus licheniformis transformation system, such as inoculum size, initial cell density in transformation medium, and time of optimal competence, have been explored, and a methodology has been developed to obtain greater reproducibility and maximal competence for transformation. The transformation system in B. licheniformis has been shown to be similar to that of B. subt...
